抑制CAR-T细胞衰竭的策略研究

摘要: 免疫治疗在临床抗肿瘤治疗中取得了良好的疗效,尤其是嵌合抗原受体T细胞(CAR-T)疗法作为一种新兴的肿瘤免疫疗法,在血液系统恶性肿瘤临床治疗中取得了突破性的疗效。但CAR-T细胞在治疗过程中会面临逐渐衰竭的问题,特别是在实体瘤的治疗中,导致抗肿瘤疗效欠佳及复发等风险。文章分析了CAR-T细胞衰竭的可能机制,并阐述了抑制CAR-T细胞衰竭的相关研究进展,以期设计合理的治疗策略,减少CAR-T细胞的衰竭,提高CAR-T细胞的效能。Abstract: Immunotherapy has achieved good efficacy in clinical anti-tumor therapy, especially chimeric antigen receptor T cell (CAR-T cell) therapy, as an emerging tumor immunotherapy, has achieved breakthrough efficacy in the clinical treatment of hematologic malignancies. However, CAR-T cell face the problem of gradual exhaustion during the treatment process, especially in the treatment of solid tumors, which will lead to poor anti-tumor efficacy and possible risks such as recurrence. This paper analyzed the possible mechanism of CAR-T cell exhaustion and describes the relevant research progress in inhibiting CAR-T cell exhaustion, with a view to designing reasonable therapeutic strategies to reduce CAR-T cell exhaustion and improve the efficacy of CAR-T cell.
