川芎嗪对人宫颈癌HeLa细胞增殖的影响及其分子机制

  • 摘要: 目的:研究川芎嗪对人宫颈癌HeLa细胞增殖的影响,并探讨其与趋化因子CXCR4表达水平之间的关系。方法:体外培养人宫颈癌HeLa细胞,经川芎嗪作用48 h后,应用MTT法检测川芎嗪对HeLa细胞增殖活性的抑制,然后采用流式细胞仪检测经川芎嗪处理的实验组与以磷酸盐缓冲液处理的对照组中HeLa细胞的细胞周期分布;应用蛋白印迹法半定量及免疫荧光实验定性检测川芎嗪处理后趋化因子受体CXCR4表达水平的改变。结果:川芎嗪对人宫颈癌HeLa细胞的增殖有明显的抑制作用,与对照组相比差异具有统计学意义(P<0.01)。经川芎嗪处理后的HeLa细胞G0/G1期增多,S期细胞减少,与对照组比较差异有统计学意义(P<0.01)。川芎嗪能够下调人宫颈癌HeLa细胞的CXCR4蛋白表达。结论:川芎嗪对人宫颈癌细胞HeLa的增殖有明显抑制作用,其抑制作用可能与下调HeLa细胞中CXCR4蛋白的表达有关。

     

    Abstract: Objective:To observe the effect of tetrame-thylpyrazine (TMP) on cell proliferation and cell cycle phase of human cervical carcinoma cell line HeLa and to elucidate its relationship with CXCR4 expression.Methods:HeLa cells were treated with TMP for 48 hours. Methylthiazolyl tetrazolium assay was performed to reveal the inhibitory effect on cell proliferation, and Flow Cytometry was used to determine the cell cycle of the HeLa cell line. Western blot and immunofluorescence were adopted to evaluate the expression of CXCR4 on the HeLa cells.Results:We found that TMP remarkably inhibited cell viability and growth of HeLa cells by MTT(P<0.01).Flow cytometry demonstrated that TMP increased the rate of G0/G1 of HeLa cell cycle(P<0.01).Western blot and immunofluorescence results showed that TMP down-regulated the expression of CXCR4 in HeLa cells. Conclusion: TMP can inhibit the viability and growth of human cervical carcinoma HeLa cell line. Our results indicate that TMP-mediated treatment involve inhibiting the expression of chemokine CXCR4 in HeLa cells.
