基因挖掘软件GenCLip寻找强直性脊柱炎相关基因的研究

  • 摘要: 目的:以AS为例,分析探讨基因挖掘软件GenCLip用于寻找与疾病相关的基因的可行性。方法:根据全基因组cDNA芯片结果,在常规统计方法初筛得出的25个差异基因(强直性脊柱炎与健康志愿者比较)的基础上,采用GenCLip寻找与AS相关的基因。结果:在用2个关键词Fibrosis、Inflammation创建网络时发现了2个新的与AS相关的基因RGS1和CCL3L3,它们和AS的炎症、纤维化网络相关,通过实时荧光定量法证实RGS1是一个未分化脊柱关节病重要的生物标记,而CCL3L3在AS中的作用则有待进一步研究。结论:用GenCLip软件分析芯片结果寻找疾病相关基因可能是一个新的可行方法,建议推广应用以进一步验证其可靠性。

     

    Abstract: Objective:Take Ankylosing spondylitis(AS) as an example to approach the manipulation and feasibility of GenCLip—a gene mining software in searching disease-related genes. Methods:According to the results from genomewide cDNNA chips,use GenCLip to search AS disease-related genes among genes prescreened.Results:Using two keywords “fibrosis” and “inflammation”,author set a network and found two related genes with ankylosing spondylitis. The two new genes are RGS1 and CCL3L3. We have confirmed that the RGS is an important biological marker in undifferentiated spondyloarthropathy, and the effect of CCL3L3 in AS remains to be studied further. Conclusions:GenCLip—a gene mining software may be a value and new mathod to find related disease gene based on the gene array data,its dependability should be validated.
