Dectin-1、CARD9基因在角质形成细胞对红色毛癣菌应答中的作用

The roles of Dectin-1 and CARD9 genes in the response of HaCaT cells against Trichophyton rubrum

  • 摘要: 目的 研究树突状细胞相关性C型凝集素-1(Dectin-1)和胱天蛋白酶募集域蛋白9(CARD9)基因在角质形成细胞对红色毛癣菌应答中的作用。 方法 将红色毛癣菌与正常HaCaT细胞、Dectin-1、CARD9基因敲除HaCaT细胞共培养,用ELISA分析其细胞上清液中IL-6、 IL-8的表达情况。 结果 Dectin-1基因敲除导致HaCaT细胞IL-6和IL-8基础分泌明显增加,敲除CARD9基因导致HaCaT细胞IL-6基础分泌增加(P < 0.05)。在红色毛癣菌的刺激下,和未感染相比,HaCaT细胞分泌的IL-8明显减少(P < 0.05);Dectin-1基因敲除的HaCaT细胞则表现为IL-6、IL-8分泌量明显减少(P均< 0.05);CARD9基因敲除的HaCaT细胞的IL-6、IL-8分泌则无明显变化(P均> 0.05)。 结论 Dectin-1基因和CARD9基因可能参与了维持表皮免疫稳态,CARD9基因还可能介导了红色毛癣菌引起的免疫应答。

     

    Abstract: Objective To investigate the roles of Dectin-1 and caspase recruitment domain-containing protein9 (CARD9) genes in the response of HaCaT cells against Trichophyton rubrum. Methods In this experiment, Trichophyton rubrum was co-cultured with HaCaT cells, Dectin-1-/- HaCaT cells and CARD9-/- HaCaT cells, respectively. The expression levels of IL-6 and IL-8 in the cell supernatant were assessed by ELISA. Results Dectin-1 gene knockout resulted in a significant increase in the secretion of IL-6 and IL-8(both P < 0.05), CARD9 gene knockout increased the secretion of IL-6 in HaCaT cells(P < 0.05). After the infection of Trichophyton rubrum, the secretion of IL-8 was significantly decreased in HaCaT cells(P < 0.05). Dectin-1-/- HaCaT cells showed a significant decrease of IL-6 and IL-8(both P < 0.05), whereas no evident change was observed in the secretion of IL-6 and IL-8 in CARD9-/- HaCaT cells(both P > 0.05). Conclusions Dectin-1 and CARD9 genes may play an important role in maintaining the immune homeostasis of the epidermis. CARD9 gene may be involved in mediating the immune response against Trichophyton rubrum.
